SECTION A – Short Answer Section
This section contains short conceptual questions related to microbial classification, sterilization parameters, microbial spoilage, and laboratory techniques used in pharmaceutical microbiology. These questions test the basic knowledge of microorganisms and their role in pharmaceutical science.
Question 1: What are Prokaryotes and Eukaryotes? Give one example of each.
Answer:
Prokaryotes:
Microorganisms that do not have a true nucleus or membrane-bound organelles.
Example: Bacteria (Escherichia coli).
Eukaryotes:
Organisms that contain a true nucleus and membrane-bound organelles.
Example: Fungi or yeast.
Question 2: Difference between Antiseptic and Disinfectant.
Answer:

Used for biological and pharmaceutical research.
SECTION B – Long Answer Section
This section requires detailed explanations of sterilization methods, bacterial structure, and cell culture techniques used in microbiology laboratories.
Question 1: Explain Physical Methods of Sterilization.
Answer:
Physical sterilization methods destroy microorganisms using heat, radiation, or filtration.
1. Moist Heat Sterilization:
Uses steam under pressure in an autoclave.
Standard condition: 121°C for 15 minutes.
2. Dry Heat Sterilization:
Uses hot air oven at 160–170°C for sterilization.
3. Filtration:
Removes microorganisms using membrane filters.
4. Radiation:
UV radiation for surface sterilization.
Gamma radiation for medical equipment.
Question 2: Explain Ultra Structure of Bacteria.
Answer:
The bacterial cell structure includes:
Cell wall: Protects the cell.
Cell membrane: Controls transport of substances.
Cytoplasm: Contains enzymes and nutrients.
Nucleoid: Genetic material (DNA).
Flagella: Helps in movement.
Capsule: Provides protection.
Bacteria are classified morphologically into:

Question 3: Explain Animal Cell Culture and its Applications.
Answer:
Animal cell culture is the process of growing animal cells in laboratory conditions.
Steps:
Tissue extraction

Applications:

Production of monoclonal antibodies.
SECTION C – Descriptive Section
This section tests deeper knowledge of microbial contamination, virus cultivation, sterility testing, and microbiological assays.
Question 1: Why do we need to Preserve Microbes? Explain Two Methods.
Answer:
Microbes are preserved to maintain pure cultures for research and industrial use.
Methods of preservation:
Refrigeration Method:
Microorganisms are stored at low temperatures.
Lyophilization (Freeze Drying):
Microbes are frozen and dried under vacuum for long-term storage.
Question 2: Write about Gram Staining.
Answer:
Gram staining differentiates bacteria into Gram-positive and Gram-negative bacteria.
Steps:
Crystal violet staining
Iodine treatment
Alcohol decolorization
Safranin counterstaining
Results:
Gram-positive bacteria → Purple
Gram-negative bacteria → Pink
Question 3: Define Virus and Methods of Cultivation.
Answer:
A virus is a submicroscopic infectious agent that can replicate only inside a host cell.
Methods of virus cultivation:
Animal inoculation
Embryonated egg method
Tissue or cell culture technique
These methods help study viruses and develop vaccines.
